I really should talk about how ELISPOT assays are conducted sometime. First though, I think I'll rant about its purpose, and later, its advantages.

The enzyme-linked ImmunoSpot assay is a potent tool for detecting and analyzing individual cells that secrete a particular protein in vitro, and for enumerating them. This technique was originally developed for analyzing specific antibody-secreting B lymphocytes, but has since been adapted for measuring the frequencies of cells that produce and secrete cytokines and a variety of other effector molecules.
